Divergence between Active in situ Degraders and Enriched Isolates Shown by Stable-isotope Based Fatty Acid Analysis

چکیده
The characterization of polutant-degrading microorganisms from contaminated soils, waste water treatment plants or biofilters for the cleaning of waste gases usually starts with enrichment cultures and ends with the taxonomical and functional characterization of the isolated strains. However, the use of cultivation independent techniques, like cloning and sequencing of marker genes, indicate the presence of a high number of still unknown microorganisms with unknown functions. It is still not clear to what extent the active populations of these complex systems are properly represented by the isolated degraders and how much of the existing biodegradation potential is still not accessable by the available isolation techniques.

Stable Isotope-based in-situ Labelling as a Reference for the Isolation of relevant Styrene Degraders from Biofilters
The enrichment of styrene degrading microorganisms from biofilters resulted in a broad range of degrading isolates with unkown in situ relevance. For this reason an in situ labeling of styrene degrading microorganisms using deuterated styrene and phospholipid fatty acids as biomarkers prior to the isolation approach was performed. By this method the fatty acid profiles of the primary styrene de...
متن کاملField-based stable isotope probing reveals the identities of benzoic acid-metabolizing microorganisms and their in situ growth in agricultural soil.
We used a combination of stable isotope probing (SIP), gas chromatography-mass spectrometry-based respiration, isolation/cultivation, and quantitative PCR procedures to discover the identity and in situ growth of soil microorganisms that metabolize benzoic acid. We added [(13)C]benzoic acid or [(12)C]benzoic acid (100 microg) once, four times, or five times at 2-day intervals to agricultural fi...

P48 protein of Mycoplasma agalactiae is used to diagnose infection and was identified as potential vaccine candidate. According to the genetic nature of mycoplasma and variable sensitivity in P48-based serological diagnosis tests, intra species variation of P48 nucleotide sequence investigated in 13 field isolates of difference province of Iran along with three vaccine strains. Samples were col...
متن کاملStable Isotope Probing Identifies Novel m-Xylene Degraders in Soil Microcosms from Contaminated and Uncontaminated Sites
The remediation of groundwater contaminated with benzene, toluene, ethylbenzene, and the xylenes (BTEX) typically involves in situ biodegradation. Although the mechanisms of aerobic BTEX biodegradation in laboratory cultures have been well studied, less is known about the microorganisms responsible in mixed culture samples or at contaminated sites. In this study, the microorganisms responsible ...
